Spanish champions CR La Vila will make their European debut when they host former finalists and French outfit Agen at the Estadio de Rugby on Saturday afternoon.
The Division De Honor winners will be taking their first bow in any European competition when they face the 1998 finalists and three time semi-finalists this weekend.
Agen lost 43-5 to Colomiers back then but will be looking to kick off their new Amlin Challenge Cup campaign with a victory over Spain's only representative in Europe.
CR La Vila coach Mark Hewitt, who spent three years in charge at the Cornish Pirates, will know his side will have to be on their mettle against a side currently lie in sixth in the Top 14.

Sale Sharks, the former Amlin Challenge Cup winners, travel to France to face former Heineken Cup winners, Brive, in a clash of the champions at the Stadium Municipal.
The English side, who won the tournament in 2002 and 2005, will be hoping to end a two-match losing streak in their domestic competition to record a victory over one of the iconic sides in European rugby history.
Brive, who lifted the Heineken Cup in 1996, will take part in their 11th season of Amlin Challenge Cup rugby but the double semi-finalists will be looking to go one step further in the competition.
Steve Diamond's Sale side, who recently announced the signing of Scottish star Richie Gray for next season, will be determined to come away from France with a major scalp as they bid for a third title.
