Stade Français Paris have guaranteed their place in the Top 16 play-offs with a 27-16 victory over European Shield finalists Auch.
The Paris club, who decided not to return to the French capital after their game against Brive on Friday, but spent the time in the south of France, were losing at half-time but came back to bag the four points.
However, Toulouse decided to risk their play-off position by resting virtually their entire first team for their visit to Agen. The only first team players on duty were Benoit Baby and David Gerard, who is returning from injury.
Toulouse lost 55-11 against a full strength Agen team who are vying for seventh place in the Top 16 and potentially the 7th Heineken Cup qualification if Pau are unsuccessful in the European Challenge Cup final against Sale Sharks at the Kassam Stadium in Oxford on 21 May. Agen scored eight tries in Toulouse's heaviest defeat of the season.
The results leave Castres Olympique in fifth place, just five points behind Toulouse and that precious play-off position, with the clubs clashing on Saturday in a key match.
The Toulouse game was dedicated to the memory of 25-year-old Maher Khala, an Agen player who also spent two seasons at Toulouse who lost his battle with stomach cancer last weekend.
